Description:
Glucovance combines two antidiabetic agents: metformin (a biguanide) and glibenclamide (a sulfonylurea), used to manage blood glucose in patients with type 2 diabetes mellitus. Metformin decreases hepatic glucose production and enhances insulin sensitivity, while glibenclamide stimulates insulin secretion from the pancreas.
Indications:
For the treatment of type 2 diabetes mellitus in adults when diet, exercise, and monotherapy with either metformin or glibenclamide do not provide adequate glycemic control.
Dosage and Administration:
Starting dose: Usually one tablet once or twice daily with meals to reduce gastrointestinal side effects.
Dosage should be individualized based on blood glucose response.
Do not exceed the maximum recommended daily dose (usually 2000 mg metformin / 20 mg glibenclamide).
Tablets should be swallowed whole with water.
Contraindications:
Type 1 diabetes mellitus
Diabetic ketoacidosis
Moderate to severe renal or hepatic impairment
Known hypersensitivity to sulfonylureas, metformin, or any tablet component
Pregnancy and lactation
Warnings and Precautions:
Risk of lactic acidosis from metformin, especially with impaired renal function
Risk of hypoglycemia, particularly in elderly or those with irregular meals
Monitor blood glucose and renal function regularly
Use caution with alcohol intake
